The Human BAFF-R Enzyme-Linked Immunosorbent Assay (ELISA) tools (ab213839) is designed for the quantitative measurement of Human TNFRSF13C in cell custom supernatants, cell lysates, serum and plasma (heparin, EDTA).

The ELISA tools is based on commonplace sandwich enzyme-linked immune-sorbent assay experience. A monoclonal antibody from mouse explicit for BAFF-R has been pre-coated onto 96-well plates. Necessities (Expression system for regular: NSO; Immunogen sequence: S7-A71) and check out samples are added to the wells, a biotinylated detection polyclonal antibody from goat explicit for BAFF-R is added subsequently after which adopted by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Sophisticated is added and unbound conjugates are washed away with PBS or TBS buffer. HRP substrate TMB is used to visualise HRP enzymatic response. TMB is catalyzed by HRP to produce a blue shade product that turned yellow after together with acidic stop reply. The density of yellow is proportional to the Human BAFF-R amount of sample captured in plate.

  • Notes

BAFF-R (additionally known as Tumor necrosis situation receptor superfamily member 13C or B-cell-activating situation receptor)  is a protein that in folks is encoded by the TNFRSF13C gene. By homology to a BAC clone, the BAFFR gene was mapped to chromosome 22q13.1-q13.31. B-cell activating situation (BAFF) enhances B-cell survival in vitro and is a regulator of the peripheral B-cell inhabitants. The protein encoded by this gene is a receptor for BAFF and is a sort III transmembrane protein containing a single extracellular phenylalanine-rich space. It is thought that this receptor is the principal receptor required for BAFF-mediated mature B-cell survival.

Description: Tumor necrosis factor receptor superfamily member 13C (TNFRSF13C), also known as BAFFR, is a protein in humans is encoded by the TNFRSF13C gene. The BAFFR gene is mapped to chromosome 22q13.1-q13.31. It has got 184 amino acid transmembrane protein which is 56% identical to the mouse protein. B cell-activating factor (BAFF) enhances B-cell survival in vitro and is a regulator of the peripheral B-cell population. BAFF plays a crucial role in B cell development and can function through receptors other than BCMA.

Recombinant Human BAFF-R (TNFRSF13C)

Z100015 50 µg
EUR 105
Description: The B cell-activating factor from the TNF family (BAFF), is emerging as an important regulator of B cell and T cell responses. BAFF was originally identified as a factor responsible for B cell survival and maturation. BAFF binds to several receptors. These include transmembrane activator and calcium modulator and cyclophilin ligand interactor (TACI), BAFF-R (BR3), and B cell maturation Ag (BCMA). BAFF-R appears to be particularly important for the regulation of B cell survival and maturation in the spleen, because A/WySnJ mice expressing a defective BAFF-R have disrupted B cell maturation, similar to that seen in BAFF-deficient mice.

Description: BAFF is mainly produced by innate immune cells such as neutrophils, monocytes, macrophages, dendritic cells, follicular dendritic cells. T cells, activated B cells, some malignant B cells and also non-lymphoid cells like astrocytes, synoviocytes and epithelial cells can also produce BAFF. BAFF binds three distinct receptors (BAFF-R, TACI and BCMA) expressed predominantly on B cells, although activated T cells also express BAFF-R. BAFF is a master regulator of peripheral B cell survival, and together with IL-6, promotes Ig class-switching and plasma cell differentiation . Besides its major role in B cell biology, BAFF co-stimulates activated T cells. Deregulated expression of BAFF leads to autoimmune disorders in mice. In humans, elevated levels of soluble BAFF have been detected in the serum of patients with various autoimmune diseases, such as Sjögren’s syndrome , Rheumatoid Arthritis (RA) , Multiple sclerosis (MS) and Systemic Lupus Erythematosus (SLE) . BAFF is also increased levels in some lymphoid cancers.
